Supercritical fluid chromatography (SFC) has traditionally been viewed as a powerful tool for chiral separations. Recent research shows a surge in SFC being used for achiral applications, particularly in the pharmaceutical sector. This talk will explore the theory behind SFC, emphasizing its advantages over HPLC, including:
- SFC is a Green technology: Reduced organic solvent consumption
- Faster separations: Lower viscosity CO2 can be used at higher flow rates
- Broad applicability: Effective for both chiral and achiral molecules, and offers orthogonal selectivity and separation
We will delve into key components for SFC method development, such as modifier selection, column temperature, back pressure, and flow rate optimization. Additionally, we will showcase novel applications of SFC beyond chiral separations, including analysis of lipids, small metabolites, food nutrients, and environmental contaminants.
We will compare SFC and HPLC systems, and demonstrate SFC's compatibility with various detectors and common packed columns. Finally, the presentation will emphasize the ease of use of SFC, making it a readily accessible technique for chromatographers familiar with HPLC.
